That depends. According to information that I have gotten, some 250's had the 200 bellhousing pattern and most had the 302 pattern. Go to www.wubba.net and there you will find a FAQ about putting an AOD on an old mustang, can't remembewr if it was a 302 or I6. Problem is the tranny mounts are Mav specials and the 302 and I6 have different mounts. Putting in another tranny might mean fabricating a tranny mount. Also, depending on the Tranny, you might have to bang out the tunnel a little and that causes headaches with bucket seats. Finally, with "Modern" trannies you need a computer that was never on a Mav, Tranny+PC+custom mount+tranny tunnel body work = $$$$. Could try an old world class T-5, though.
